The catalytic addition reactions of ferrocenyl ketones with terminal olefins in the presence of Ru(H)2(CO)(PPh3)3 as catalyst have been studied. Benzoylferrocene reacts with triethoxyvinylsilane, styrene and vinylferrocene, respectively, to give 1:1 coupling products I-III in high yields. C-H bond cleavage takes place at the carbon atom of the benzene ring at the ortho position of the carbonyl group and C-C bond formation takes place at the terminal carbon atom of the olefins. 2-Furoylferrocene reacts with vinylferrocene to give a 1:1 coupling product IV and the C-H bond cleavage takes place at the carbon atom of the furan ring at the ortho position of the carbonyl group and the C-C bond formation takes place at the terminal carbon atom of vinylferrocene. The new products I-IV have been characterized by elemental analysis, 1H-NMR and MS. The X-ray crystal structure of IV has been determined.

Amperometric biosensor for hypoxanthine based on immobilized xanthine oxidase on nanocrystal gold-carbon paste electrodes

The preparation and performance of a xanthine oxidase (XOD) biosensor, based on a carbon paste electrode (CPE) modified with electrodeposited gold nanoparticles (nAu), for the amperometric determination of hypoxanthine (Hx) is reported. Different XOD biosensor configurations were evaluated and compared with electrodes constructed by immobilizing XOD onto unmodified CPE and with biosensors prepared using glassy carbon electrodes and gold disk electrodes modified with electrodeposited gold. The XOD-nAu-CPE in which the enzyme was immobilized by cross-linking with glutaraldehyde (GA) and BSA exhibited the highest amperometric signal for Hx. Although Hx detection is usually carried out at potential values of around +600 mV versus Ag/AgCl, the GA-BSA-XOD-nAu-CPE allowed this detection to be carried out at 0.00 V, thus minimizing potential interferences from electrochemically oxidizable substances such as ascorbic acid. Experimental variables concerning the biosensor preparation were optimized. Calibration plots for Hx were constructed with the biosensor operating at +600 mV and at 0.00 V. The detection limit for Hx, 2.2 ¡Á 10-7 mol l-1, obtained using the latter potential value is similar to the best detection limits reported in the literature with other biosensor designs working at much more extreme potentials. The usefulness of the biosensor for the analysis of real samples was demonstrated by determining Hx in sardines and chicken meat.

Syntheses and properties of multiferrocenylated corannulenes

The syntheses and properties of corannulenes bearing different numbers and types of ferrocenyl groups are described. Six different monoferrocenylated corannulenes were synthesized, and the crystal structure of 1-corannulenyl-1?-(ferrocenyl)benzene was elucidated by single-crystal X-ray analysis. Further, diferrocenylated corannulenes bearing methyl or trifluoromethyl groups are reported. Buckybowls with four and five ferrocenyl substituents were synthesized from tetrabromocorannulene and the symmetrical pentachlorocorannulene. The molecular structure and nutshell-like crystal packing of a tetraferrocenylated corannulene was determined by single-crystal X-ray analysis. Functional and structural characterization of an ECF-type ABC transporter for vitamin B12

Vitamin B12 (cobalamin) is the most complex B-type vitamin and is synthetized exclusively in a limited number of prokaryotes. Its biologically active variants contain rare organometallic bonds, which are used by enzymes in a variety of central metabolic pathways such as L-methionine synthesis and ribonucleotide reduction. Although its biosynthesis and role as co-factor are well understood, knowledge about uptake of cobalamin by prokaryotic auxotrophs is scarce. Here, we characterize a cobalamin-specific ECF-type ABC transporter from Lactobacillus delbrueckii, ECF-CbrT, and demonstrate that it mediates the specific, ATP-dependent uptake of cobalamin. We solved the crystal structure of ECF-CbrT in an apo conformation to 3.4 A resolution. Comparison with the ECF transporter for folate (ECF-FolT2) from the same organism, reveals how the identical ECF module adjusts to interact with the different substrate binding proteins FolT2 and CbrT. ECF-CbrT is unrelated to the well-characterized B12 transporter BtuCDF, but their biochemical features indicate functional convergence.
