We have synthesized a novel disubslituted ferrocenyl compound [Fe(C 5H4CO(CH=CH)2C6H4NEt 2)2] (3) that displays a remarkable fluorescence quantum yield (1.1 ¡Á 10-1) in acetonitrile, and we have studied its capacity for calcium detection in depth using both electrochemical and optical techniques in this medium. The results of our NMR analysis reveal that the ligand-calcium interaction is CO-centered and that an uncommon equilibrium occurs between 3 and calcium triflate, involving five species of different stoichiometries. In contrast, our analysis of the UV-vis absorption data indicates that only three species of different stoichiometries are formed when calcium perchlorate is used with 3. Mass spectrometry measurements provide strong support for the formation of all these different species in solution. In addition, the electrochemical detection of calcium triflate by 3 leads to an irreversible FeII/ FeIII oxidation process with an unusual negative shift (-60 mV) caused by the nBu4NBF4 salt effect on the Ca2+-3 interaction process. Compound 3 can also be an original optical probe to detect calcium perchlorate over a wide range of salt concentration by UV-vis absorption spectroscopy. The most original and intriguing property of compound 3 is that it exhibits an unprecedented “multistep” fluorescence behavior upon addition of this salt.

Synthesis and physico-analytical studies of some novel ferrocenyl Schiff base derivatives

A series of ferrocenyl Schiff base derivatives was synthesized by condensation reactions of 1,1?-ferrocenedicarboxaldehyde and aromatic amines containing long chain alkyl groups as free ends which were characterized by their physical properties, elemental, FTIR, 1H NMR, 13C NMR spectral and thermal analysis. The thermal behaviour of the synthesized compounds was studied by differential scanning calorimetry (DSC) which revealed that these compounds may exhibit mesomorphic properties. The DSC results of aromatic amines and ferrocenyl Schiff bases were compared to study the effects of structure, i.e. rigid core and terminal chain length, on the phase transition behaviour.

Rosmarinic acid is a novel inhibitor for hepatitis b virus replication targeting viral epsilon RNA-polymerase interaction

Current therapeutics for hepatitis B virus (HBV) patients such as nucleoside analogs (NAs) are effective; however, new antiviral drugs against HBV are still desired. Since the interaction between the epsilon (epsilon) sequence of HBV pregenomic RNA and viral polymerase (Pol) is a key step in the HBV replication cycle, we aimed to identify small compounds for its inhibition, and established a pull-down assay system for the detection of epsilon-RNA-binding-Pol. Screening showed that 5 out of 3,965 compounds inhibited epsilon-Pol binding, and we identified rosmarinic acid, which exhibited specificity, as a potential antiviral agent. In order to examine the anti-HBV effects of rosmarinic acid, HBV-infected primary human hepatocytes from a humanized mouse liver were treated with rosmarinic acid. The rosmarinic acid treatment decreased HBV components including the amounts of extracellular HBV DNA with negligible cytotoxicity. We also investigated the combined effects of rosmarinic acid and the NA, lamivudine. rosmarinic acid slightly enhanced the anti-HBV activity of lamivudine, suggesting that the HBV replication step targeted by rosmarinic acid is distinct from that of NA. We analyzed an additional 25 rosmarinic acid derivatives, and found that 5 also inhibited epsilon-Pol. Structural comparisons between these derivatives implied that the ?two phenolic hydroxyl groups at both ends? and the ?caffeic acid-like structure? of rosmarinic acid are critical for the inhibition of epsilon-Pol binding. Collectively, our results demonstrate that rosmarinic acid inhibits HBV replication in HBV-infected cells by specifically targeting epsilon-Pol binding.
