Comparison study of live cells by atomic force microscopy, confocal microscopy, and scanning electrochemical microscopy

In this report, three kinds of scanning probe microscopy techniques, atomic force microscopy (AFM), confocal microscopy (CM), and scanning electrochemical microscopy (SECM), were used to study live cells in the physiological environment. Two model cell lines, CV-1 and COS-7, were studied. Time-lapse images were obtained with both contact and tapping mode AFM techniques. Cells were more easily scratched or moved by contact mode AFM than by tapping mode AFM. Detailed surface structures such as filamentous structures on the cell membrane can be obtained and easily discerned with tapping mode AFM. The toxicity of ferrocenemethanol (Fc) on live cells was studied by CM in reflection mode by recording the time-lapse images of controlled live cells and live cells with different Fc concentrations. No significant change in the morphology of cells was caused by Fc. Cells were imaged by SECM with Fc as the mediator at a biased potential of 0.35 V (vs. Ag/AgCl with a saturated KC1 solution). Cells did not change visibly within 1 h, which indicated that SECM was a noninvasive technique and thus has a unique advantage for the study of soft cells, since the electrode scanned above the cells instead of in contact with them. Reactive oxygen species (ROS) generated by the cells were detected and images based on these chemical species were obtained. It is demonstrated that SECM can provide not only the topographical images but also the images related to the chemical or biochemical species released by the live cells.

Direct inkjet printing of a complete and working amperometric biosensor for the detection of hydrogen peroxide, based on horseradish peroxidase (HRP), has been demonstrated. This result paves the way for an extensive application of “biopolytronics”, i.e. the utilization of conductive/semiconductive polymers and biologically active molecules to design bioelectronic devices using a common PC, and exploiting normal commercial printers to print them out.

Solution viscosity effects on the heterogeneous electron transfer kinetics of ferrocenemethanol in dimethyl sulfoxide-water mixtures

The electrochemical kinetics for the oxidation of ferrocenemethanol (FcCH2OH) over the whole composition range of dimethyl sulfoxide (DMSO)-water solutions of different viscosities (I¡¤) containing 50.0 mM (CH3)4NClO4 (TMAP) at a Pt microelectrode was studied using scanning electrochemical microscopy (SECM). The measured diffusion coefficient, DFcCH2OH, as well as the standard rate constant of the heterogeneous electron transfer, k0, as a function of solution composition, showed a minimum at about a DMSO molar fraction (xDMSO) of 0.33, corresponding to the mixture with the maximum solution viscosity. The largest k0 value found, 2.06 A¡À 0.31 cm s-1 in pure water (electrolyte) medium, was about 15 times larger than that obtained in the solution of xDMSO = 0.33 (0.14 A¡À 0.02 cm s-1). A good linear correlation between ln k0 and ln I¡¤ was observed within the solution composition range of 0.10 a?? xDMSO a?? 0.60. An excellent linear correlation between ln k0 and ln I?L, the longitudinal relaxation time, was also obtained with a slope equal to 1.0 when xDMSO = 0-0.60. Unusually small rate constants found in the solutions of xDMSO a?? 0.70 were attributed to adsorption effects at the tip and the substrate electrode. The k0 obtained for the present system was generally found to be inversely proportional to the viscosity of the solution and directly proportional to the diffusion coefficient of the electroactive species.

Synthesis, chemical reactivity and electrochemical behaviour of mono- and difluoro metallocenes

We report the synthesis of mono- and 1,1?-difluoro-substituted metallocenes (ferrocene, ruthenocene) and of asymmetrical 1,1?-disubstituted ferrocenes with one substituent being fluorine. Lithiation of metallocenes and subsequent addition of the fluorinating agent NFSI gave the fluorinated metallocenes after optimization of the experimental conditions. All new compounds were comprehensively characterized and the cyclic voltammograms of fluoro- and 1,1?-difluoroferrocene were recorded and compared to other mono- and dihalogenated ferrocenes. Half-wave potentials of +106 mV and +220 mV vs. FcH0/+ were obtained for monofluorinated species and difluorinated ferrocene, respectively. Both values are remarkably low compared to the other halogenated ferrocenes (Cl, Br, and I). Finally, 1-bromo-1′-fluoro-ferrocene turns out to be an ideal starting material for further fluoro-substituted ferrocene derivatives.
