Neuroblastoma (NB) is an embryonic malignancy affecting the physiological development of adrenal medulla and paravertebral sympathetic ganglia in early infancy. Proteasome inhibitors (PIs) (i.e., carfilzomib (CFZ)) may represent a possible pharmacological treatment for solid tumors including NB. In the present study, we tested the effect of a novel non-competitive inhibitor of heme oxygenase-1 (HO-1), LS1/71, as a possible adjuvant therapy for the efficacy of CFZ in neuroblastoma cells. Our results showed that CFZ increased both HO-1 gene expression (about 18-fold) and HO activity (about 8-fold), following activation of the ER stress pathway. The involvement of HO-1 in CFZ-mediated cytotoxicity was further confirmed by the protective effect of pharmacological induction of HO-1, significantly attenuating cytotoxicity. In addition, HO-1 selective inhibition by a specific siRNA increased the cytotoxic effect following CFZ treatment in NB whereas SnMP, a competitive pharmacological inhibitor of HO, showed no changes in cytotoxicity. Our data suggest that treatment with CFZ produces ER stress in NB without activation of CHOP-mediated apoptosis, whereas co-treatment with CFZ and LS1/71 led to apoptosis activation and CHOP expression induction. In conclusion, our study showed that treatment with the non-competitive inhibitor of HO-1, LS1 / 71, increased cytotoxicity mediated by CFZ, triggering apoptosis following ER stress activation. These results suggest that PIs may represent a possible pharmacological treatment for solid tumors and that HO-1 inhibition may represent a possible strategy to overcome chemoresistance and increase the efficacy of chemotherapic regimens.

Abstract A series of 1,1′-di(hydroxyalkyl)ferrocenes, [Fc'{(CH 2)nOH}2], with n = 1 (1), 2 (2), 3 (3) and 4 (4) and Fc’ = Fe(eta5-C5H4)2, was synthesized. The electrochemistry of the di(hydroxyalkyl)ferrocenes was studied by cyclic voltammetry in CH2Cl2/0.1 M [N nBu4][PF6] utilizing a glassy carbon working electrode. The ferrocenyl group showed reversible electrochemistry with the formal reduction potential, Eo’ , inversely proportional to alkyl chain length and approximately 59 mV smaller than those of the corresponding mono(hydroxyalkyl)ferrocenes derivatives [Fc(CH2)mOH] with m = 1 (1m), 2 (2m), 3 (3m), and 4 (4m) and Fc = Fe(eta5-C 5H5)(eta5-C5H4 -). The tetraalcohol [Fc'{CH(OH)(CH2)3OH} 2], 5, possessing four OH functionalities, two in the terminal positions and two more, one on each of the two alpha-C relative to the ferrocenyl (Fc’ for dialcohols or Fc for monosubstituted derivatives) group, was isolated as a side product during the synthesis of 4. The formal reduction potential of 5 was Eo’ = -24 mV vs. FcH/FcH+ and closely approached Eo’ of [FcCH(OH)CH3] (Eo’ = -11 mV), [Fc'{CH(OH)CH3}2] (-21 mV) and 1 (0.00 mV vs. FcH/ FcH+). The single crystal X-ray structure of the tetraalcohol 5 (Z = 8, orthorhombic, space group Pbca) was also solved.

Aims: H2O2 is produced by all eukaryotic cells under physiological and pathological conditions. Due to its enormous relevance for cell signaling at low concentrations and antipathogenic function at high concentrations, precise quantification of extracellular local hydrogen peroxide concentrations ([H2O2]) originating from single cells is required. Results: Using a scanning electrochemical microscope and bare platinum disk ultramicroelectrodes, we established sensitive long-term measurements of extracellular [H2O2] kinetics originating from single primary human monocytes (MCs) ex vivo. For the electrochemical techniques square wave voltammetry, cyclic and linear scan voltammetry, and chronoamperometry, detection limits for [H2O2] were determined to be 5, 50, and 500 nM, respectively. Following phorbol ester stimulation, local [H2O2] 5-8 mum above a single MC increased by 3.4 nM/s within the first 10 min before reaching a plateau. After extracellular addition of H2O2 to an unstimulated MC, the local [H2O2] decreased on average by 4.2 nM/s due to degradation processes of the cell. Using the scanning mode of the setup, we found that H2O2 is evenly distributed around the producing cell and can still be detected up to 30 mum away from the cell. The electrochemical single-cell measurements were validated in MC populations using electron spin resonance spectroscopy and the Amplex UltraRed assay. Innovation and Conclusion: We demonstrate a highly sensitive, spatially, and temporally resolved electrochemical approach to monitor dynamics of production and degradation processes for H2O2 separately. Local extracellular [H2O2] kinetics originating from single cells is quantified in real time.
