A chemobiocatalytic strategy for the highly stereoselective synthesis of nitrile-substituted cyclopropanes is reported. The present approach relies on an asymmetric olefin cyclopropanation reaction catalyzed by an engineered myoglobin in the presence of ex situ generated diazoacetonitrile within a compartmentalized reaction system. This method enabled the efficient transformation of a broad range of olefin substrates at a preparative scale with up to 99.9 % de and ee and up to 5600 turnovers. The enzymatic product could be further elaborated to afford a variety of functionalized chiral cyclopropanes. This work expands the range of synthetically valuable, abiotic transformations accessible through biocatalysis and paves the way to the practical and safe exploitation of diazoacetonitrile in biocatalytic carbene transfer reactions.

The iron(III) protoporphyrin IX complex with imidazole, a biologically relevant ligand, occupying an axial position, has been studied by infrared multiple photon dissociation (IRMPD) spectroscopy. The complex has been delivered in gas-phase by electrospray ionization (ESI), mass selected in an ion trap, and assayed by IRMPD spectroscopy in two complementary frequency regions. The fingerprint range (900-1900 cm-1) has been scanned using the Orsay free-electron laser beamline (CLIO), while the X-H (X = C,N,O) stretching region (3000-3600 cm-1) has been inspected using a tabletop IR optical parametric oscillator/amplifier (OPO/OPA) laser source. DFT calculations have been performed to obtain a comprehensive pattern of the various potential conformers yielding optimized geometries, relative thermodynamic parameters, and respective IR spectra. The comparison between the IR spectra for representative conformers and the experimental IRMPD features suggests the coexistence of two families of conformers involving different degrees of folding and hydrogen bonding between the two propionic acid functionalities on the periphery of the protoporphyrin IX macrocycle in a ratio depending on environmental conditions such as ESI solvent and temperature. The observed conformational variability of the porphyrin substituents in the naked heme-imidazole complex is consistent with the fine-tuning of the reactivity properties of this important prosthetic group by the specific surroundings in the protein core.

MF6p/FhHDM-1 is a small protein secreted by the parasitic flatworm (trematode) Fasciola hepatica that belongs to a broad family of heme-binding proteins (MF6p/helminth defense molecules (HDMs)). MF6p/HDMs are of interest for understanding heme homeostasis in trematodes and as potential targets for the development of new flukicides. Moreover, interest in these molecules has also increased because of their immunomodulatory properties. Here we have extended our previous findings on the mechanism of MF6p/HDM-heme interactions and mapped the protein regions required for heme binding and for other biological functions. Our data revealed that MF6p/FhHDM-1 forms high-molecular-weight complexes when associated with heme and that these complexes are reorganized by a stacking procedure to form fibril-like and granular nanostructures. Furthermore, we showed that MF6p/FhHDM-1 is a transitory hemebinding protein as protein-heme complexes can be disrupted by contact with an apoprotein (e.g. apomyoglobin) with higher affinity for heme. We also demonstrated that (i) the heme-binding region is located in the MF6p/FhHDM-1 C-terminal moiety, which also inhibits the peroxidase-like activity of heme, and (ii) MF6p/HDMs from other trematodes, such as Opisthorchis viverrini and Paragonimus westermani, also bind heme. Finally, we observed that the N-terminal, but not the C-terminal, moiety of MF6p/HDMs has a predicted structural analogy with cell-penetrating peptides and that both the entire protein and the peptide corresponding to the N-terminal moiety of MF6p/FhHDM-1 interact in vitro with cell membranes in hemin-preconditioned erythrocytes. Our findings suggest that MF6p/HDMs can transport heme in trematodes and thereby shield the parasite from the harmful effects of heme.

We extend our approach of combination chemotherapy through a single prodrug entity (O’Neill et al. Angew. Chem., Int. Ed. 2004, 43, 4193) by using a 1,2,4-trioxolane as a protease inhibitor carbonyl-masking group. These molecules are designed to target the malaria parasite through two independent mechanisms of action: iron(II) decomposition releases the carbonyl protease inhibitor and potentially cytotoxic C-radical species in tandem. Using a proposed target “heme”, we also demonstrate heme alkylation/carbonyl inhibitor release and quantitatively measure endoperoxide turnover in parasitized red blood cells.

We recently reported that exposure to cyclosporine (CSA) plus diclofenac causes hypertension and impairs left ventricular (LV) and cardiac autonomic functions in female rats. Here, we tested the hypothesis that these effects could be mitigated by facilitated heme oxygenase (HO) signaling. Experiments were performed in female rats to assess the effects of 10-day treatment with CSA (25 mg/kg/day)/diclofenac (1 mg/kg/day) regimen on cardiovascular functions in absence and presence of maneuvers that upregulate HO or its enzymatic products. The CSA/diclofenac-induced hypertension and impairment in cardiac sympathovagal balance (i.e. reduced low-frequency/high-frequency spectral ratio) were blunted upon concurrent treatment with hemin (HO-1 inducer), tricarbonyldichlororuthenium (II) dimer (CORM-2, carbon monoxide-releasing molecule), or bilirubin. While none of the latter treatments affected the CSA/diclofenac-evoked decrease in isovolumic relaxation constant (Tau, a measure of diastolic function), the increased LV contractility (dP/dtmax) and attenuated reflex bradycardia in CSA/diclofenac-treated rats was abolished by bilirubin only. Paradoxically, the CSA/diclofenac-evoked attenuation in reflex tachycardia was improved in presence of hemin or CORM-2, but not bilirubin. The favorable hemin effects were abrogated after inhibition of HO (ZnPP) or nitric oxide synthase (NOS, L-NAME). These finding highlights NOS-dependent modulatory roles for HO and its enzymatic products in improving the worsened cardiovascular profile in CSA/diclofenac-treated female rats.

We synthesized a series of hemin derivatives (HDs) substituted by residues of amino acids and peptides at either one or two propionic-acid residues, and studied the virucidal activity of the compounds obtained against herpes simplex virus. Compounds 6,7-bis-(methyl ester N0 L-seryl)-protohemin (IX) (2) and 6,7-bis-[methyl ester N0-L-arginyl)-protohemin (IX) (6) shown the highest virucidal activity. We also investigated the interaction between HDs and lipid-membrane components as a possible mechanism of virucidal action. A model system including Clark’s electrode and a micellar solution of methyl linoleate was used to quantitatively assess the capability of HDs to catalyze the oxidation of polyunsaturated fatty acids as components of lipid membranes. Another model system including liposomes that consisted of dioleoylphosphatydylcholine and was loaded with the fluorescent dye carboxyfluorescein was employed to examine the effect of HDs on lipid-membrane permeability. The kinetics and efficacy of increasing liposome-membrane permeability on exposure to HDs appeared to depend on the nature of the substituents in the HDs. The findings are strongly suggestive of the presence of two different modes of interaction between an HD and the lipid membrane, i.e. oxidative and non-oxidative mechanisms possibly underlie the virucidal action of HDs.

Current therapeutics for hepatitis B virus (HBV) patients such as nucleoside analogs (NAs) are effective; however, new antiviral drugs against HBV are still desired. Since the interaction between the epsilon (epsilon) sequence of HBV pregenomic RNA and viral polymerase (Pol) is a key step in the HBV replication cycle, we aimed to identify small compounds for its inhibition, and established a pull-down assay system for the detection of epsilon-RNA-binding-Pol. Screening showed that 5 out of 3,965 compounds inhibited epsilon-Pol binding, and we identified rosmarinic acid, which exhibited specificity, as a potential antiviral agent. In order to examine the anti-HBV effects of rosmarinic acid, HBV-infected primary human hepatocytes from a humanized mouse liver were treated with rosmarinic acid. The rosmarinic acid treatment decreased HBV components including the amounts of extracellular HBV DNA with negligible cytotoxicity. We also investigated the combined effects of rosmarinic acid and the NA, lamivudine. rosmarinic acid slightly enhanced the anti-HBV activity of lamivudine, suggesting that the HBV replication step targeted by rosmarinic acid is distinct from that of NA. We analyzed an additional 25 rosmarinic acid derivatives, and found that 5 also inhibited epsilon-Pol. Structural comparisons between these derivatives implied that the ?two phenolic hydroxyl groups at both ends? and the ?caffeic acid-like structure? of rosmarinic acid are critical for the inhibition of epsilon-Pol binding. Collectively, our results demonstrate that rosmarinic acid inhibits HBV replication in HBV-infected cells by specifically targeting epsilon-Pol binding.

We have probed single-molecule redox reaction dynamics of hemin (chloride) adsorbed on Ag nanoparticle surfaces by single-molecule surface-enhanced Raman spectroscopy (SMSERS) combined with spectroelectrochemistry. Redox reaction at the molecule/Ag interface is identified and probed by the prominent fluctuations of the Raman frequency of a specific vibrationalmode, ?4, which is a typical marker of the redox state of the iron center in a hemin molecule. On the basis of the autocorrelation and crosscorrelation analysis of the single-molecule Raman spectral trajectories and the control measurements of single-molecule spectroelectochemistry and electrochemical STM, we suggest that the single-molecule redox reaction dynamics at the hemin-Ag interface is primarily driven by thermal fluctuations. The spontaneous fluctuation dynamics of the single-molecule redox reaction is measured under no external electric potential across the molecule-metal interfaces, which provides a novel and unique approach to characterize the interfacial electron transfer at the molecule-metal interfaces. Our demonstrated approaches are powerful for obtaining molecular coupling and dynamics involved in interfacial electron transfer processes. The new information obtained is critical for a further understanding, design, and manipulation of the charge transfer processes at the molecule-metal interface or metal-molecule-metal junctions, which are fundamental elements in single-molecule electronics, catalysis, and solar energy conversion.
